According to your post, my understanding is that you got 401 unathourized error.
You d better to provide the scenario how you got the error.
If you get the 401 unauthorized error when you access the site home page, you need to make sure you set the site URL as an alternate access mapping for this web application.
If you get the 401 unauthorized error when your app tries to access SharePoint content, you need to go into IIS and find your provider hosted app, then disable Anonymous access and enable Windows authentication. I recommend restarting the IIS virtual
server for your hosted app, then try again.
In addition, the error may also caused by the IIS authenticate, the custom master page, IIS binding and so on.
