Do EMET could possibly blocked Botnet or possibly mitigate Botnet attack?

EMET is not a detection tool, it is a mitigation tool.

There are possible scenarios were EMET would alert you on malicious behavior caused by some malware/botnet software, but it's intended goal is rather to make sure you cannot get infected by the malware.
