In the initial configuration of a newly installed SCCM site server which is also a Software distribution point, I went to the Component Configuration and Software Distribution Point. In the general tab, there is a chance to specify the "location of stored packages". If I fill in with F:\, does it mean all packages created in the future will be stored in F:\ drive? Thanks.

Yes. But this isn't talking about the source file location as specified on each package or the actual distribution point location.
