 What do i need to check if automatic reply is not working for all of our users? We setup correctly the automatic reply but still not working.

1. Make sure that database is mounted. If no, please mount it and check the result.

2. Run the Test-AssistantHealth cmdlet to verify that the Microsoft Exchange Mailbox Assistants service is healthy.

Have you checked OOF is enabled? If yes, check smart host setting. However, I'm not sure it works, but you should give it a try as described here

Your question asked how to know if it's working, not how to configure it.  Automatic replies to the Internet are configured using Set-RemoteDomain.
