Which part of the procedure do you need help with? Is it to create it in the first place or add users to that mailbox store?
.
There is a pretty decent guide that explains it short but fairly accurate here

.
If you need more help, please feel free to let me know

Your subject line has 'Mailbox', but the question itself has 'Mailbox Store'. You may not be aware of the difference. The word Store in this context refers to a database containing several mailboxes. So, just to clear things up, do you want to create a
new Mailbox or an actual Store?Mobile OWA For Smartphone

You mean in the New Mailbox wizard, on the Mailbox Settings page, underneath where it says 'Specify the mailbox database rather than using a database automatically selected'? That's normally blank.Mobile OWA For Smartphone

You need to create your users in EMC now that you have Exchange 2010. In the Recipient Configuration part, choose New Mailbox. That will create a new Mailbox-enabled User Account. The only reason you would now create users in AD, is if
you didn't want them to have a mailbox. Using EMC in E2010 allows you to create the user and the mailbox at the same time.Mobile OWA For Smartphone

If you don't mind, i will add a few details.
As Lee said, you should create your users from the EMC, this was both the case in exchange 2007 and 2010, however, if you have a user that already exists, and want to move this account from one mailbox store to another, you can find the user in EMC, rightclick
on the user in question and choose "move mailbox".
If you have a user that has been created in AD but don't have a mailbox, you can go to the EMC, under recipient configuration, choose new mailbox, choose user mailbox, now you should be asked if you wish to create a mailbox for a user that already exists
or if you wish to create a new user, choose the user that already exists, find the user you wish to give a mailbox and finish the wizard.
If you wish to create a new user, go to the EMC, under recipient configuration, choose new mailbox, fill out the details, on the next page, you can choose the users Alias and what mailbox store it should use, hit the browse button and choose the newly created
database and finish it.Yours Faithfully Martin
