I want to upgrade exchange server 2010 to Exchange 2010 SP1 but i have some confusion Client Access and Hub Transport roll are installed one 1 server and Mailbox are installed to another system first which server i can upgrade

You should upgrade to SP2 ( and RU2) instead.
