I currently have a spanned volume with a database on it, on a Windows Server 2003.

If I create a new simple volume with the required space and copy over the files from the spanned volume, then delete the spanned volume and give the spanned volume drive letter to the newly created simple volume, should everything run as normal?

I think you directly copy the files from the spanned volume to the new simple volume with the same drive letter. But if you want to retain the same NTFS permission, I suggest you use robocopy to achieve:
