If ur using 32 bit of win2k8 it is not possible. u have to go for clean installation. R2 is 64 bit. If 2008 is 64 bit, you can refer the to the article :Upgrade Domain Controllers and also thread provided by Santhosh.Gopi Kiran |Facebook| This posting is provided AS IS with no warranties, and confers no rights.

pls refer the below mentioned information(copied from technet).if you have a 2k8 64 bit server you can upgrade your server. In-place upgrades from Windows Server 2003 or Windows Server 2003 R2 to Windows Server 2008 or Windows Server 2008 R2 are supported (subject to supportedWindows Server 2008 R2 Upgrade Paths), with the following exception: x86-based operating systems cannot be upgraded in place to x64-based versions of Windows Server 2008 or Windows Server 2008 R2 (which supports only the x64-based architecture). An x64-based version of Windows Server 2008 can be upgraded in-place to Windows Server 2008 R2. A writeable domain controller cannot be upgraded to be an RODC. The reverse is also true. A server that runs the full installation of Windows Server 2008 R2 cannot be upgraded to be a server that runs a Server Core installation of Windows Server 2008 R2. The reverse is also true. The best practice for introducing domain controllers that run a newer operating system than current domain controllers is to make the new domain controller an additional domain controller in an existing domain. For more information about supported and unsupported upgrades, seeWindows Server 2008 R2 Upgrade Paths(http://go.microsoft.com/fwlink/?LinkID=154894). Windows Server 2008 and Windows Server 2008 R2 both auto-install Internet Protocol version 6 (IPv6). Do not arbitrarily disable or remove IPv6. Darshana Jayathilake
