In the documentation on this page on step 3.1 when enabling Load Balancing for DA: https://technet.microsoft.com/en-us/library/jj134209.aspx there is an option to change the IPv6 Prefix.

1. In the middle pane of the console, in the Step 2 DirectAccess Server area, click Edit.

2. Click Prefix Configuration.

On my Win2012R2 server setup With DA with a single NIC, I don't have that option. What is wrong here?

This is only when the DirectAccess server has an IPv6 static address.

You do not need to worry about this if you are using IPv4 only on the DirectAccess server.
